Schematic representation of the insulin-like growth factor (IGF) system. The IGF system consists of IGF-I and IGF-II, IGF binding proteins (IGFBP-1 to7), and type I and II IGF receptors. IGFBPs have high affinity to IGFs and modulate their access to IGF receptors. IGF proteases dissociate the IGFBP-IGF complex, and the freed IGFs can then bind to the IGF receptors, activating downstream signaling pathways: the phosphatidylinositol-3-kinase and RAS-extracellular signal-regulated kinase pathways.
