Effects of dexamethasone pretreatment on chemotherapy-induced cytotoxicity. (a) Dexamethasone pretreatment (10-7 M) inhibits chemotherapy-induced cytotoxicity in the MCF-7 cell line. (b) However, clusterin antisense oligonucleotides (ODNclu) increase chemotherapy-induced cytotoxicity in the presence of dexamethasone. Cells were treated with dexamethasone for 4 hours. After dexamethasone treatment, the medium was replaced in some wells with medium containing 10-7 M paclitaxel or doxorubicin for 1 day. Cell viability was determined by trypan blue exclusion test. Each data point represents the mean percentages of at least three independent experiments ± standard error of the mean. *p < 0.05; **p < 0.01; ***p < 0.001.
